    Window Query Processing with Proxy Cache

    A location dependent query (LDQ) result set is valid only in a specific region called the validity region (VR). While limiting the validity of a particular result set to a given area, the VR may also be used in caching implementations to determine if cached results satisfy semantically equivalent queries. Existing LDQ caching schemes rely on the database servers to provide the VR at a cost of high computational overhead. Alternatively, a LDQ proxy cache, which approximates the VR can be employed, freeing the database servers from the high cost of calculating the VR. A LDQ proxy cache architecture is proposed to compute an estimated validity region (EVR) based on the observed querying history at the proxy server. We present an algorithm - Window_EVR - for the LDQ proxy to compute the EVR for a window query result set. The simulation results show that LDQ proxy caching using the Window_EVR algorithm significantly reduces both the window query response time and the workload at the database servers while maintaining query result set accuracy

    Transactional Agents for Pervasive Computing

    Pervasive computing enables seamless integration of computing technology into everyday life to make upto- date information and services proactively available to the users based on their needs and behaviors. We aim to develop a transaction management scheme as a pertinent component for such environment supported by either structured or ad hoc networks. We propose Transactional Agents for Pervasive COmputing (TAPCO), which utilizes a dynamic hierarchical meta data structure that captures the semantic contents of the underlying heterogeneous data sources. Mobile agents process the transactions collaboratively, to preserve ACID properties without violating local autonomy of the data sources. TAPCO is simulated and compared against Decentralized Serialization Graph Testing (DSGT) protocol. The results show that TAPCO outperforms DSGT in several ways. In contrast to DSGT that did not consider local transactions, TAPCO supports both local and global transactions without violating the local autonomy

    Multimedia Correlation Analysis in Unstructured Peer-to-Peer Network

    Recent years saw the rapid development of peer-topeer (P2P) networks in a great variety of applications. However, similarity-based k-nearest-neighbor retrieval (k-NN) is still a challenging task in P2P networks due to the multiple constraints such as the dynamic topologies and the unpredictable data updates. Caching is an attractive solution that reduces network traffic and hence could remedy the technological constraints of P2P networks. However, traditional caching techniques have some major shortcomings that make them unsuitable for similarity search, such as the lack of semantic locality representation and the rigidness of exact matching on data objects. To facilitate the efficient similarity search, we propose semantic-aware caching scheme (SAC) in this paper. The proposed scheme is hierarchy-free, fully dynamic, non-flooding, and do not add much system overhead. By exploring the content distribution, SAC drastically reduces the cost of similarity-based k-NN retrieval in P2P networks. The performance of SAC is evaluated through simulation study and compared against several search schemes as advanced in the literature

    Association of germline genetic variants with breast cancer-specific survival in patient subgroups defined by clinic-pathological variables related to tumor biology and type of systemic treatment

    BACKGROUND: Given the high heterogeneity among breast tumors, associations between common germline genetic variants and survival that may exist within specific subgroups could go undetected in an unstratified set of breast cancer patients. METHODS: We performed genome-wide association analyses within 15 subgroups of breast cancer patients based on prognostic factors, including hormone receptors, tumor grade, age, and type of systemic treatment. Analyses were based on 91,686 female patients of European ancestry from the Breast Cancer Association Consortium, including 7531 breast cancer-specific deaths over a median follow-up of 8.1 years. Cox regression was used to assess associations of common germline variants with 15-year and 5-year breast cancer-specific survival. We assessed the probability of these associations being true positives via the Bayesian false discovery probability (BFDP < 0.15). RESULTS: Evidence of associations with breast cancer-specific survival was observed in three patient subgroups, with variant rs5934618 in patients with grade 3 tumors (15-year-hazard ratio (HR) [95% confidence interval (CI)] 1.32 [1.20, 1.45], P = 1.4E-08, BFDP = 0.01, per G allele); variant rs4679741 in patients with ER-positive tumors treated with endocrine therapy (15-year-HR [95% CI] 1.18 [1.11, 1.26], P = 1.6E-07, BFDP = 0.09, per G allele); variants rs1106333 (15-year-HR [95% CI] 1.68 [1.39,2.03], P = 5.6E-08, BFDP = 0.12, per A allele) and rs78754389 (5-year-HR [95% CI] 1.79 [1.46,2.20], P = 1.7E-08, BFDP = 0.07, per A allele), in patients with ER-negative tumors treated with chemotherapy. CONCLUSIONS: We found evidence of four loci associated with breast cancer-specific survival within three patient subgroups. There was limited evidence for the existence of associations in other patient subgroups. However, the power for many subgroups is limited due to the low number of events. Even so, our results suggest that the impact of common germline genetic variants on breast cancer-specific survival might be limited

    Silencing Dkk1 expression rescues dexamethasone-induced suppression of primary human osteoblast differentiation

    <p>Abstract</p> <p>Background</p> <p>The Wnt/β-catenin pathway is a major signaling cascade in bone biology, playing a key role in bone development and remodeling. The objectives of this study were firstly, to determine the effects of dexamethasone exposure on Wnt/β-catenin signaling at an intracellular and transcriptional level, and secondly, to assess the phenotypic effects of silencing the Wnt antagonist, Dickkopf-1 (Dkk1) in the setting of dexamethasone exposure.</p> <p>Methods</p> <p>Primary human osteoblasts were exposed in vitro to 10<sup>-8 </sup>M dexamethasone over a 72 h time course. The phenotypic marker of osteoblast differentiation was analyzed was alkaline phosphatase activity. Intracellular β-catenin trafficking was assessed using immunoflourescence staining and TCF/LEF mediated transcription was analyzed using a Wnt luciferase reporter assay. Dkk1 expression was silenced using small interfering RNA (siRNA).</p> <p>Results</p> <p>Primary human osteoblasts exposed to dexamethasone displayed a significant reductions in alkaline phosphatase activity over a 72 h time course. Immunoflourescence analaysis of β-catenin localization demonstrated a significant reduction in intracytosolic and intranuclear β-catenin in response to dexamethasone exposure. These changes were associated with a reduction of TCF/LEF mediated transcription. Silencing Dkk1 expression in primary human osteoblasts exposed to dexamethasone resulted in an increase in alkaline phosphatase activity when compared to scrambled control.</p> <p>Conclusions</p> <p>Wnt/β-catenin signaling plays a key role in regulating glucocorticoid-induced osteoporosis <it>in vitro</it>. Silencing Dkk1 expression rescues dexamethasone-induced suppression of primary human osteoblast differentiation. Targeting of the Wnt/β-catenin signaling pathway offers an exciting opportunity to develop novel anabolic bone agents to treat osteoporosis and disorders of bone mass.</p

    Gene expression analysis in human osteoblasts exposed to dexamethasone identifies altered developmental pathways as putative drivers of osteoporosis

    BACKGROUND: Osteoporosis, a disease of decreased bone mineral density represents a significant and growing burden in the western world. Aging population structure and therapeutic use of glucocorticoids have contributed in no small way to the increase in the incidence of this disease. Despite substantial investigative efforts over the last number of years the exact molecular mechanism underpinning the initiation and progression of osteoporosis remain to be elucidated. This has meant that no significant advances in therapeutic strategies have emerged, with joint replacement surgery being the mainstay of treatment. METHODS: In this study we have used an integrated genomics profiling and computational biology based strategy to identify the key osteoblast genes and gene clusters whose expression is altered in response to dexamethasone exposure. Primary human osteoblasts were exposed to dexamethasone in vitro and microarray based transcriptome profiling completed. RESULTS: These studies identified approximately 500 osteoblast genes whose expression was altered. Functional characterization of the transcriptome identified developmental networks as being reactivated with 106 development associated genes found to be differentially regulated. Pathway reconstruction revealed coordinate alteration of members of the WNT signaling pathway, including frizzled-2, frizzled-7, DKK1 and WNT5B, whose differential expression in this setting was confirmed by real time PCR. CONCLUSION: The WNT pathway is a key regulator of skeletogenesis as well as differentiation of bone cells. Reactivation of this pathway may lead to altered osteoblast activity resulting in decreased bone mineral density, the pathological hallmark of osteoporosis. The data herein lend weight to the hypothesis that alterations in developmental pathways drive the initiation and progression of osteoporosis
